El partido de Lanús forma parte de un importante conglomerado urbano en el cordón del territorio que rodea a la ciudad capital, conocido como "Gran Buenos Aires"​. Limita con los municipios de Lomas de Zamora, Quilmes, Avellaneda y la Ciudad de Buenos Aires (Capital Federal) de la cual está separada por una verdadera barrera geográfica: el Riachuelo. La vía de conexión directa con la capital es el Puente Alsina. La superficie total de Lanús es de 48,35 Km2, siendo uno de los más pequeños municipios del Gran Buenos Aires pero contando con una mayor densidad de población que los otros por kilómetro cuadrado. Las tierras del Partido de Lanús integran en casi su totalidad, la cuenca hidrográfica del Río Matanza, modesto afluente del Río de la Plata, pero eje de un extenso sistema de arroyos y lagunas por cuyo curso originalmente serpenteante, corría en un lecho de barro y tosca, de poca profundidad, a través de un suelo aluvional, anegadizo y salitroso. Este ha sido en épocas prehistóricas un brazo del mar que ocupaba todo el estuario del Plata, que cubría el actual municipio de Avellaneda, la mayor parte de Lanús, amplias zonas de Capital Federal"​. Esta característica salitrosa ha quedado registrada, entre otras cosas, por los depósitos salitrosos y abundantes, moluscos marinos, y la capa de arena y limo muy fino que recubre el estrato de tosca en las zonas aledañas al río de La Matanza"​. "Sobre ella se ha detectado otra capa con restos de tortugas, moluscos y peces de agua dulce, indicando que después de retirarse el mar del estuario rioplatense, las aguas dulces en avance, ocupaban también sectores poblados del Partido de Lanús y zonas limítrofes.
